I just wanted to know if i can start a litte Let's Play "Amnesia - A Machine for Pigs" once its released.
I need a licence, but the Friction Team does not reply...

You don't need a license to make Amnesia Let's Plays.
But if YouTube has denied it, maybe you're using a copyrighted material.

False proposition. Amnesia
is copyrighted work, just like any other piece of work that someone creates. The difference is between the fact that if you upload this material somewhere, are the copyright holders going to hunt you down for using their material.
In the case of Amnesia let's plays: No. Frictional Games don't mind.
For crying out loud, we have a separate section dedicated to Amnesia/Penumbra gameplays. This already should answer the question.

Little advice from someone who had to deal with Let's Play fine print.
You can play the game the SECOND it comes available, however, if you want to be safe from having your channel being hit with a copyright strike, start posting the game the second it releases in Los Angeles. This was a rule that was passed onto me by Machinima and I recommend you follow that rule if you want to avoid any problems
